FOXO4-DRI (also known as Proxofim) is a synthetic D-retro-inverso peptide designed as a potent senolytic agent. It targets senescent ("zombie") cells — dysfunctional cells that accumulate with age and contribute to inflammation, tissue dysfunction, and age-related decline — while sparing healthy cells.
FOXO4-DRI works by disrupting the interaction between the transcription factor FOXO4 and p53 protein in senescent cells. This interference promotes p53 nuclear exclusion, triggering selective apoptosis (programmed cell death) in these damaged cells without broadly affecting healthy tissue.
