PT-141, also known as Bremelanotide, is a synthetic peptide analog of α-MSH primarily studied for its effects on sexual function and libido in both male and female research models. Unlike traditional treatments that primarily influence vascular blood flow, PT-141 acts centrally through the nervous system.
Mechanistically, PT-141 functions as a melanocortin receptor agonist with highest affinity for MC4R receptors in the brain. Activation of these receptors enhances dopamine signaling in key areas such as the hypothalamus, promoting sexual motivation, arousal, and desire. It also modulates related pathways involving mood, stress response, and appetite. Because of its central mechanism of action, PT-141’s effects in laboratory studies are largely independent of vascular function, distinguishing it from PDE5 inhibitors.
